Misterstraat continues on from Landstraat in Bredevoort and begins near the bridge over the Boven-Slinge, where the Misterpoort once stood.
The road once served as the eastern route towards the Winterswijk hamlet of Miste and formed part of the Hessian road between Aalten and Winterswijk.
